Antonine started on the front foot with Fraser James having a shot saved. The same player then hit the post as Antonine piled on the pressure before Lewis Smith saw a couple of efforts saved by the Port keeper.
After 20 minutes, a Danny Hamilton corner was not cleared and Chris Donaghy fired home to make it 1-0.10 minutes later a Steven MacIver cross was headed goalwards by Smith but it was blocked on the line by a defender.
Two minutes before the break a long ball found the Port striker, who had beaten the offside trap and he made it 1-1 against the run of play.
Shortly after the restart, a Donaghy cross was flicked on by Dom Cairney and Hamilton smashed home a tremendous volley to give Antonine a deserved lead.
Three minutes later, a Port Glasgow striker broke free of the defence and headed over Andy Carroll to make it 2-2.
On the hour mark, Smith was pulled to the ground and a penalty was awarded which Brendan Kelly calmly fired home to regain the lead.
Antonine had not finished there. A quick Scott Hutton free kick found Hamilton at the corner of the box who cut in and fired home an unstoppable shot to complete the scoring.
It was a good professional performance by Antonine in game they controlled from start to finish.
